The Journel of the Jesuits mentioned that the Council of the Order announced that it had unanimously approved of Guillaume's marriage. It was on November 28, 1649 that he married Anne Esmard (Aymard). She was baptized on October 22, 1627, in Saint André de Niort, Poitou. She was the daughter of the late Jean and Marie Bineau. Anne had two sisters in Canada: Barbe, wife of Gilles Michel dit Taillon, and after him, of Oliver Letardif; and Madeleine, wife of Zacharie Cloutier. The wedding of Guillaume and Anne took place in the house of Couture, at Pointe Lévy, in the presence of Father Jean LeSeur, Chaplain of the Hospitalliers of Québec. The couple engendered ten children: 6 boys and 4 girls.

Elle a immigré en Nouvelle-France en 1648 avec ses deux soeurs, Barbe et Marie-Madeleine, qui épousèrent respectivement Olivier LeTardif et Zacharie Cloutier. Vers 1648, abjuration d'Anne Émard, elle a renoncé à sa religion protestante calviniste et a été baptisé catholique pour épouser, à 22 ans, Guillaume Couture, 30 ans.. Étaient présents au mariage dans la demeure du couple: Olivier LeTardif, Zacharie Cloutier, Martin Gravel, Charles Cadieu. Jean Le Sueur a célébré la cérémonie de mariage. Le couple aura 10 enfants. Elle est décédée à 72 ans, le 17 janvier 1700 et ses funérailles ont eu lieu le 18 janvier 1700. Son corps fut inhumé dans le cimetière de la paroisse Notre-Dame à St. Joseph de Lévis à Québec, Nouvelle-France.
